Rampant Hearts surged eight ahead of their Glasgow rivals at the top of the Scottish top flight to strengthen their early championship claims and deepen the struggles around Brendan Rodgers' side.
Alexandros Kyziridis' fine strike and Lawrence Shankland's penalty gave the Hearts team a 3-1 lead that the title holders could not recover from.
The young defender - one of three youngsters in the Celtic XI - scored an own goal in the early stages to put Hearts ahead at a bouncing Tynecastle, and it was the youngster's infringement that led to the penalty.
Callum McGregor's classy first goal of the season had quickly cancelled out Hearts' opener, and Benjamin Nygren almost put Celtic in front, opting for finesse over power and allowing Alexander Schwolow to save from close range.
But the unstoppable Hearts side seized control again to consign Celtic to back-to-back losses for the first time since last year.
It also gives the Hearts a significant lead at the top of the table.
The last time they were so far clear was in 2005-06 under their former manager, when they ended as runners-up - a rare instance that a team has failed to win the title after building such an advantage in the last three decades.
McInnes, whose former team were second to Celtic on four occasions, has been working hard to play down championship discussions, as the club seek their first top-flight crown since 1960.
The Celtic boss has experienced this previously at the club - both flawless and stuttering starts to a season. He's yet to finish second best.
But on Sunday's evidence, this Edinburgh outfit showed they deserve serious consideration.
Match Analysis: Hearts Capitalize of Weakened Celtic
Hearts started with the self-assurance of a side unbeaten in the league this season - fans' favourite Kyziridis delivered a ball for Shankland to stab wide.
The Glasgow team didn't aid their cause with some unusual sloppiness in possession and they buckled under their initial test.
A headed effort from Braga was tipped onto the woodwork by Kasper Schmeichel and Shankland's return ball was diverted goalwards by the unfortunate Murray, in for the injured Cameron Carter-Vickers.
Celtic grew into the contest and started stringing passes together. From a flowing attack, Sebastian Tounekti released Tierney to set up the captain, who slotted home.
Apart from the opportunity for Nygren, Celtic found it hard to generate chances for striker Kenny or his replacement Shin Yamada amid the unavailability of stricken attackers Kelechi Iheanacho and Daizen Maeda.
Hearts rediscovered their energy and adaptability in creating chances in the opposition half. Kyziridis, a standout performer of the club's scouting network, fired a low shot past Schmeichel from the edge of the box.
Almost straight away, the defender tripped the midfielder in the right side of the box and Shankland scored. And it could've been an even more handsome win, had substitute Ageu not been stopped by Schmeichel.
The visitors' performance after the break was partially encapsulated in a couple of moments involving record signing Arne Engles.
The set-piece from Engles delivery was in stark contrast to his midweek exploits in the continental victory over their Austrian opponents and the Belgium international was then booked for a professional foul.
Post-Match Comments
Hearts head coach Derek McInnes: "You see how hard the players work, the togetherness. That's not sufficient on its own to win games consistently.
"Our strength lies in, is skillful players. You're witnessing it from a lot of players who have been here before. Recent additions are eager to show that.
"We've got a good thing going at the minute, but it's just one win. It's important for us to keep winning. We've got much work ahead, loads to do. Following the initial fixtures is normally an indication of your standing."
The Celtic boss: "The second goal was disappointing from our perspective. We didn't move when the play was recycled as a line so when they pass the ball, they should have players offside but weren't. We have got to do better with the attempt.
"Plenty of matches to play for. It's only the beginning. It's only nine games in. Obviously, being eight adrift is not ideal. The positive is there's numerous fixtures to play.
"We now have to focus on the present and look to find more consistency and more quality in our play."
